Newly created fly to study how a normal cell turns cancerous

ScienceDaily (Nov. 29, 2012) ? The wing of a fruit fly may hold the key to unraveling the genetic and molecular events that transform a normal cell into a cancerous one. The study, conducted on Drosophila melanogaster by scientists at the Institute for Research in Biomedicine (IRB Barcelona) and led by ICREA researcher Marco Mil?n, has reproduced each of the steps known to take place when a healthy cell turns cancerous. The researchers have thus provided an inexpensive and effective model that will allow the scientific community to scrutinize the genes and molecules involved in each step. Given that the vast majority of genes in Drosophila are conserved in mice and humans, the results obtained may also lead researchers to perform similar studies in more clinically relevant models.

Proceedings of the National Academy of Sciences (PNAS) has published the study online this week.

Argentinian scientist Andr?s Dekanty, a Juan de la Cierva researcher in the Mil?n lab and first author of the article, explains that "for the first time we have a genetic model that allows us to understand the events that take place, starting from when cells begin to accumulate genomic errors until the development of a tumour." The Mil?n team began by provoking genomic instability in a selection of cells in the fly wing. They then prevented these cells, with an aberrant number of chromosomes (aneuploidy), from succumbing to the cells' natural defense mechanisms so that they would survive. From there, they observed that the cells spread throughout the tissue, became mobile, activated the growth of adjacent cells, and degraded the basal membrane that kept them in place, allowing them to break free and even invade nearby tissues. "All of these events are things that we see in cancer. This fly model will therefore help us to identify each of the genes and molecules involved in epithelial cell detachment (delamination), motility, abnormal growth, basal cell degradation and invasion," says Mil?n.

"This study takes things one step beyond, however," say the researchers, "and opens a fundamental conceptual debate." This is the first time that these phenomena have been described in relation to genomic instability. "This has allowed us to propose something that hasn't yet been possible to study in depth and that now should be taken into serious consideration. Is genomic instability the cause of tumorigenesis?" says Mil?n.

Genomic instability, a cause of cancer?

Cells in all human cancers display considerable genomic instability. Their genomes are full of mistakes. "If we can demonstrate this direct correlation, we will have something very specific to work with to find precise targets. Aneuploid cells don't exist in healthy organisms. If we can identify what differentiates a cell with genomic instability from a normal cell, we may be able to identify specific treatments," says Dekanty.

Today, cancer therapies are aimed at slowing proliferation, or cell division. A major problem with this is that all cells -- both cancerous and healthy ones -- divide. Treatments are therefore associated with many side effects. "There isn't a treatment available that attacks only the cells with genomic instability," say the researchers. "If we can clearly differentiate one from the other, we'll hopefully be able to find drugs that target them specifically," explains Dekanty.

NFL Rumors: Is Jerry Jones Behind Les Miles To Arkansas?

Jerry Jones, owner of the NFL?s Dallas Cowboys, is now trying to ruin another team, but a least this time it?s at least a rival and not his own?team. Jones, who graduated from the University of Arkansas, is supposedly the man behind trying to get LSU Tigers coach Les Miles to the Razorbacks.

Sports By Brooks is reporting that Jones and Arkansas Athletic Director Jeff Long are the one?s?who are behind?entire process.

It?s no secret that Jones and Les Miles became very close when Miles was on the Cowboys coaching staff?from 1998-2000 and?that friendship was demonstrated when?Jones gave Miles his personal jet to fly home?to Ohio after Miles? father passed away.?It could also be the reason Miles? name popped up in the Cowboys head coaching search in 2010.

Word is that Jones would?contribute a significant amount of the $27.5 million in order to get Miles on board for the job at Arkansas.

You have to hand it to Jones, he really does want to win. Whether it?s at his alma mater or with the Dallas Cowboys, essentially nothing stops him from achieving his goal of winning. Well, except?naming a real general manager for the Cowboys.?At least this time he?s only helping the process and not?being the only one to ruin it.

If Miles, by some miracle, takes the job at Arkansas, Jones would be?partly responsible for taking down a rival at LSU and potentially building a winner at Arkansas. Just what Jerry?s always wanted, credit!

Jerry Jones, a man who needs to be in everyone?s business. Maybe if he spent more time helping fix the Cowboys, they?d have a better team.

It looks like the Yankees -- slowly but surely -- will bring back the members of its 2012 starting rotation. Which is not a bad thing. The Yankee starters were not their downfall. It was the lack of clutch hitting and putting the ball out of play.

The Bombers re-signed Hiroki Kuroda on a one-year deal for $15 million and are closing in on Andy Pettitte, one year, $11 million.

Mariano Rivera will also be back in the fold and, probably Ichiro too.

Down the road, GM Brian Cashman will most likely re-up underrated Russell Martin and postseason hero Raul Ibanez.

The bottom line?

When the 2013 season begins, the Bombers will have lost Rafael Soriano and the postseason challenged Nick Swisher.

Soriano will be replaced by Mariano and the Yanks can find a bat to replace Swisher. How about Cody Ross or making a trade for Shin-Soo Choo.

INFORMATION GATHERING

  • Death and dying requires great sensitivity.? Many older people are realistic in recognising that the end of life is relatively close, so this subject should be dealt with sensitively but not unduly secretively.
  • At all times, attempt to respect any expressed wishes of residents in relation to death or period of dying.
  • We also put into practice Advanced Care Planning which is based on the Liverpool Integrated Care Pathway for the dying patient (LCP).? This provides a template of care to guide our staff and to provide communication.? This would include :

?An initial assessment and care of the dying patient, care of the family and care after death of the patient.

This would provide appropriate information so that discussion and review of choices? between residents, their families, health and social care professionals can take place.? This would result in the drawing up of a palliative care plan which would improve care (allow discussion to avert difficulties at a later date) and enable family involvement and care to be individually focused.

  • Ensure the resident has got a Lasting Power of Attorney or a Living Will that they wish to follow.
  • We must attempt to ascertain and record any personal wishes in relation to residents eventual death.
  • We welcome the involvement of relatives and friends in the information gathering process, provided permission is sought from the resident.

?CARE PRIOR TO DEATH

?

  • We aim to provide the best physical care for a dying resident, seeking expert advice on specific needs, pain control and other disorders.? We strive to assist to provide peace of mind preparing for death with residents and their relatives.
  • Once a resident has been diagnosed and their care needs assessed and the decision taken that the individual needs palliative care, the palliative care plan would be put into practice.? This also provides specific information e.g. care provided, name of funeral director etc. and whether cremation or burial has been requested.
  • We attempt to provide a homely environment within a resident?s room, even if special equipment is required.
  • If a wish has been expressed to move to hospital/hospice, all assistance will be given and advice sought.
  • Recognise acute and often distressing emotions experienced at a time close to death and try to offer comfort and support.

  • We would (where possible) endeavour to offer comfort and support to family members and friends during this emotional time.

?

  • We actively encourage family and friends to visit where welcomed by a dying resident and try to make them as comfortable as possible.? We encourage family to stay with the resident if they wish.
  • We are also aware that hearing is one of the last senses to be lost and we are aware of talking sensitively in front of that resident and any distressing conversations would be held outside the room.
  • We respect the beliefs and spiritual needs of a dying resident and will seek expert advice when dealing with residents who subscribe to religious or other groups of which we have little experience.
  • When a resident is seriously ill or passes away, they are remembered in our monthly Home?s Communion Service, and residents committee meeting within each home.

AFTER DEATH

  • Remember that it is possible for the deceased to take a gasp of air after death, no pulse means death.

?

  • We will ensure we communicate with the Next of Kin or whoever is the main point of contact as soon as possible after the death, providing whatever help we can.
  • We will treat the deceased with the utmost respect in accordance with the wishes of the deceased?s family and friends.
  • During illness and after death we will be sensitive to the needs of the other residents.
  • We will make efforts to enable residents and staff to attend the funerals if they wish.? Of course, permission from the relatives will be sought first.

?PROCEDURE ON EXPECTED DEATH OF A RESIDENT? i.e. IF THE RESIDENT HAS BEEN SEEN BY GP WITHIN LAST 14 DAYS AND GP CONFIRMS CONDITION IS TERMINAL

  • Senior person on duty/on call should be contacted to confirm death.
  • Record time, date and circumstances of death on MAR sheet.
  • Call GP to confirm death or the registered nurse from Hill House who is qualified to do so.
  • Inform relatives, proprietors and Manager of death ? Record time on resident?s M.A.R.
  • Call chosen undertakers, ensure time is appropriate so that deceased can be discreetly removed, taking into account other residents and relatives.

  • Check on the care plan if there are any religious or cultural considerations before handling the body.

?

  • Check the deceased is clean and tidy, and laid out as flat as possible.? Open window a little.
  • Make a note or remove all jewellery before resident is removed by undertakers.
  • If own GP did not certify death, phone surgery at 9.00am next morning to inform residents GP.
  • Ensure after deceased is removed, room is left tidy.? Remove all laundry and bedding.

UNEXPECTED DEATH IF RESIDENT HAS NOT BEEN SEEN WITHIN 14 DAYS? BY GP FOR TERMINAL CONDITION

  • Record the time and circumstances of death accurately and do not move any possible evidence.? Document jewellery and clothes on deceased.
  • The deceased must remain untouched, the police should be contacted.
  • GP called to certify death who will then call the Coroner/their representative.
  • Advise relatives, proprietor and Manager of death.
  • The Coroner/representative will arrange for undertaker to remove the body.
  • If own GP did not certify, telephone surgery at 9.00am next working day to inform registered GP.
  • The staff on duty must document circumstances leading to death which may be called upon by Coroner.
  • Ensure after deceased is removed, room is left tidy.? Remove all laundry and bedding.
  • CQC must be informed of the death of a resident ? a Reg. 16 ? 20 form must be completed and sent.

ACTION OF RELATIVES

  • To make contact with chosen undertakers.
  • To telephone surgery at 9.00 am next day to confirm suitable time to collect Death Certificate.
  • To inform the home of certified cause of death at earliest possible time so they may notify the Commission.

LEGAL REQUIREMENTS

The following information must be sent to CQC within 24 hours of death:

A pretrial hearing began Tuesday in the case of Pfc. Bradley Manning, the Army private charged with spilling U.S. secrets to the website WikiLeaks. Manning supporters packed the courtroom at Fort Meade near Baltimore, many wearing black t-shirts with the word "Truth" in white lettering.

Military commanders involved in the confinement of Manning are expected to be questioned first at the hearing, which began Tuesday afternoon and is expected to last several days.

Manning's lawyers contend he was illegally punished by being locked up alone in a small cell for nearly nine months at the Marine Corps brig in Quantico, Va., where he had to sleep naked for several nights.

Judges can dismiss all charges if pretrial punishment is particularly egregious, but that rarely happens. The usual remedy is credit at sentencing for time served, said Lisa M. Windsor, a retired Army colonel and former Army judge advocate now in private practice in Washington.

In a 1956 case, U.S. v. Bayhand, a military appeals court ordered all charges dismissed against a soldier who had been forced during his pretrial confinement to do hard labor alongside a sentenced prisoner. The court ruled that the soldier had been given an illegal order.

Since then, there have been few, if any, cases in which pretrial punishment has led to dismissal of all charges. Lt. Col. Eric Carpenter, chairman of the criminal law department at the judge advocates school in Charlottesville, Va., said he couldn't find one but he couldn't say for sure that the remedy hasn't been granted.

Manning has also offered to take responsibility for the leak by pleading guilty to reduced charges. The military judge hasn't yet ruled on the offer and prosecutors have not said whether they would still pursue the charges against him.

He was kept at the Marine Corps brig from July 2010 to April 2011. The military contends the treatment at Quantico was proper, given Manning's classification as a maximum-security detainee who posed a risk of injury to himself or others. He was later moved to Fort Leavenworth, Kan., where he was re-evaluated and given a medium-security classification.

A United Nations investigator called the conditions of Manning's time at Quantico cruel, inhuman and degrading, but stopped short of calling it torture.

Outside Fort Meade, about two dozen people gathered in the rain and under umbrellas to show support for Manning. Some held a banner that said, "Support Bradley Manning." They then observed the afternoon courtroom proceedings.

The 24-year-old native of Crescent, Okla., faces possible life imprisonment if convicted of aiding the enemy, the most serious of the 22 charges.

He is accused of sending hundreds of thousands of classified Iraq and Afghanistan war logs and more than 250,000 diplomatic cables to the secret-spilling website WikiLeaks while he was working as an intelligence analyst in Baghdad in 2009 and 2010.

Of all the space objects humans have come across so far, asteroids are surely among the more anxiety-producing examples, thanks to their recurring habit of barreling past our planet at terrifying speeds and uncomfortably close proximity. However, it just may be that asteroids are our friends, a new paper suggests.
